- Extractive Spectrophotometric Methods for Determination of Esmolol Hydrochloride Using Acidic Triphenylmethane Dyes

Three simple and sensitive extractive spectrophotometric methods have been described for the assay of Esmolol hydrochloride either in pure form or in pharmaceutical formulations. The developed methods involve formation of coloured chloroform extractable ion-pair complexes of the drug with bromothymol blue (BTB), bromophenol blue (BPB) and bromocresol green (BCG) in acidic medium. The extracted complexes showed absorbances maxima at 415,412 and 412nm with BTB, BPB and BCG respectively. Beer's law is obeyed in the concentration ranges 2.5- 25μg/ml for all three dyes. The effect of concentration of dye and pH have been studied and optimized. The limits of detection and quantification have been determined for three methods. All the three methods have been validated as per the guidelines of ICH. The methods have been applied to the determination of drug in commercial tablets and results of analysis were validated statistically through recovery studies.Keywords

Three simple and sensitive extractive spectrophotometric methods have been described for the assay of Ambroxol hydrochloride either in pure form or in pharmaceutical formulations. The developed methods involve formation of coloured chloroform extractable ion-pair complexes of the drug with bromothymol blue (BTB), bromophenol blue (BPB) and bromocresol purple (BCP) in acidic medium. The extracted complexes showed absorbance maxima at 414, 412 and 407nm with use of the cited reagents, respectively. Beer's law is obeyed in the concentration ranges 2.5-25, 4.0-30 and 4.0-40μg/ml with BTB, BPB and BCP respectively. The effect of concentration of dye, pH, and interference of excipients have been studied and optimized. The limits of detection and quantification have been determined for three methods. All the three methods have been validated as per the guidelines of ICH. The methods have been applied to the determination of drug in commercial tablets and results of analysis were validated statistically through recovery studies.

The developed methods involve formation of colored chloroform extractable ion-pair complexes of the drug with bromothymol blue (BTB), bromophenol blue (BPB), bromocresol purple (BCP) and bromocresol green (BCG) in acidic medium. The extracted complexes showed absorbance maxima at 415 nm for all four methods. Beer's law is obeyed in the concentration ranges 2.5-25, 2.5-25, 2.0 - 25 and 2.5 - 25. μg/ml with BTB, BPB, BCP and BCG, respectively. Six drugs viz., Metoprolol tartrate, Torsemide, Diclofenac sodium, Chlorpheniramine, Loratadine and irbesartan were tested for the formation of charge transfer complexes with p-CA and the interaction formed a basis for quantitative determination of the drugs spectrophotometrically. CH3CN was found to be suitable for the analysis. A simple, specific, accurate and precise high performance liquid chromatographic method was developed for the determination of Amlodipine and Atorvastatin in Amlodipine besylate and Atorvastatin tablet dosage forms. A Hypersil BDS C18 5 μm (250×4.6 mm I.D) with column temperature 30°C in isocratic mode, with mobile phase containing Acetonitrile:10 mM Ammonium acetate buffer pH 4.0 (50:50) was used. The flow rate was 1.0 mL/min and effluent was monitored at 238 nm.The retention time of Amlodipine was 2.5 min and for Atorvastatin was 4.7 min. The linearity for Amlodipine and Atorvastatin was in the range 0.02 mg/mL to 0.12 mg/mL. The proposed method is accurate, precise, specific and rapid estimation of Amlodipine and Atorvastatin in Amlodipine besilate and Atorvastatin tablets.Keywords
