- Pharmocophoric Studies of Anti-Telomerase Drugs

Human Telomeres are the repetitive DNA sequences of TTAGGG/ AATCCC that protects chromosomes from the loss of essential genes. Telomeres are replicated by telomerase, a special polymerase that has own RNA template. Telomerase is universal marker of human cancer. The cancer cells have neoplastic immortalization rendered by upregulation of telomerase. Rationally, telomerase is an attractive target of cancer therapeutics. Anti-telomerase drugs are the novel enzyme inhibitors. We made a non-computational effort for the basic pharmocophoric design. It is based on the binding functions, , mode of actions, structural resemblances and correlation of reported IC50 values. Pyrimidine nucleosides, non-nucleosides, HIV-RTIs, Cartenoids, Flavonoids, synthetic chrome and phenyl derivatives were pharmocophorically studied. A chemical hybrid structure for anti-telomerase drug was proposed.

The telomere and telomerase are anti-aging biomolecules. Telomerase is an immortal enzyme. It maintains telomere length and promotes longevity. Telomerase holds future for anti-aging medicines. The anti-aging therapy has dramatic impact on public health by reducing the burden of age-related health difficulties- dementia, stroke, cancer and prolonging the quality of life. TA-65 (10-15 mg daily) is first formulation 1-2, derived from Chinese herb. Astragalus, for the activation of telomerase and maintenance of telomere healthiness. The nutritional destiny offers biochemical preventions against multiple pathologies related to telomere shortening. The life style free from the oxidative stress, anxiety, alcoholism, and spurious addictions prevent chromosomal abbreviations.

The telomeric shortening is linked to aging process. Telomerase reverses biological aging. Human body after the age of 35 years starts shrinking with reduction in number of cells. The body fights against aging, the harder it fights, the more tired it gets. The telomere is a biological clock, ticking down as we age and eventually leads to cell mortality. The life expectancy, life-span, and longevity are interrelated terms. They are basically anti-mortality and anti-aging. The maintenance of the genomic stability, DNA integrity and regulation of anti-aging cell physiology are biological foundation of immortality and anti-aging potential. The telomeric length (TL) is maintained by dynamic equilibrium between processes that lengthen and those that shorten telomeres. The cellular senescence perturbs this balance, fastens the aging and directs the mortality. The telomerase boosters and longevity enhancers natural products extend life-span and reduce the rate of biological aging. Their anti-mortality potentials and possible reasons of biological immortality which are beyond Hayflick limitation would be examined.

Aphrodisiac drugs are the pleasure – oriented pharmaceuticals and have value in sex – therapeutics. We made an attempt to apply new concept of chemobiological mimicry to aphrodisiac chemistry by comparing with endogenous, neurotransmitters and androgens. Dopamine, Norepinepherine, Serotonin and Testosterone were taken as the structural standards for rationalizing the selection of chemobiologic mimetics to design chemopharmacophoric aphrodicophore [CA]. This may be useful in imparting aphrodisiacal specificity at receptor levels and promoting bioenergetic sexual synergy.

Aphrodisiac herbs are gifted by erotic chemical structures. An ancient therepeutical wisdom utilized them in the ayurvedic formulations by rasayana technique. The thirty-six such formulations of contemporary market were studied to identify multifacet bioactions of rasayana for designing aphrodisicphoric models. Vajikaran therapy uses aphrodisiacs for enhancing sexual capacity and pleasure. Ayurvedically the aphrodisiac powers are ascribed to libidous effects of foods, spices, herbs and minerals. Two models of ayurvedicophoric aphrodisicophores are devised for the holistic longevity and psychosocial styling.

The dietary aphrodisiac molecules belonging to fruits, vegetables, nuts, spices, vitamins and micro-nutrients gift the creative expressions of love for the sexual esteem. Love is the cherished emotion and nutures the relationship. It is a chemical process. The neurochemicals and hormones are biochemical players of love making. Dopamine, norepinepherine, phenylethylamine, serotonin, estrogen, testosterone, oxytocin, vasopressin, endorphins and nerve growth factor are biomolecules of love. They define molecular basis of love. The chemistry of love in our brains is guided by DNA under the environmental influence. The chemical addiction of love has great demerit in modern arena. It keeps us reproducing, correctionpopulating, pooring and global warming. The martial and non-martial loves have their own achievements. The problems of love are addiction, boredom, polygamy adultery, incompatibility, loss of creativity, negative emotions, martial mishap, and biological precursor of poverty, population and pollution. Possibly the molecular basis of love can educate to keep the love-bonding free from the psychosocial and psychobiological adversities and tragedies.
