Simultaneous Estimation of Levofloxacin Hemihydrate and Ornidazole by High Performance Liquid Chromatography in Pharmaceutical Dosage Form

A new simple, selective, rapid, precise and economical reverse phase isocratic High Performance Liquid Chromatography (HPLC-RP) method has been developed for the simultaneous estimation of Levofloxacin hemihydrate and Ornidazole in pharmaceutical preparations using Kromacil 5 μ 100 A C18, 150 x 4.6 mm column and the mobile phase consisted of acetonitrile:water (40:60), pH adjusted to 3.0 *#177; 0.05 with dil. Orthophosphoric acid. The flow rate was kept at 1.3 ml/min. The Ultraviolet Violet (U.V.) detection was done at 302 nm. The retention times for these drugs were 1.051 min and 1.777 min, respectively. The HPLC method was validated. The present method was found to be specific. The number of theoretical plates came out to be fairly high, i.e., 1082 and 2051 for the Levofloxacin hemihydrate and Ornidazole respectively. The tailing factor came out to be 1.26 and 1.11 for Levofloxacin hemihydrate and Ornidazole respectively. The relative standard deviation or precision was calculated to be 0.5 and 0.3 for Levofloxacin hemihydrate and Ornidazole respectively. The method was tested for its robustness as the resolution came out to be 5.09. With this present method, the calibration curve was found to be linear and run time was just 3 min., which made the method rapid and economical.
